Product Details

The Investigator DIPplex Kit enables the amplification of 30 deletion/insertion polymorphisms (also known as INDELs) plus Amelogenin. This provides an equally sensitive alternative to STR- and SNP-based human identification. The kit is designed to analyze highly degraded DNA, thanks to the maximum amplicon size of just ~160 bp. The genetic analysis is calibrated with BT5 Matrix, an optimized dye technology.

Performance

The Investigator DIPplex Kit was validated and evaluated using the GeneAmp 9700 thermal cycler (in standard mode), and the ABI PRISM 310, ABI PRISM 3100, and Applied Biosystems 3130 Genetic Analyzers. 

The recommended amount of DNA under standard conditions is 0.35 ng. Internal validations demonstrated robust and balanced results with 0.2–0.5 ng, and reliable results with <0.1 ng DNA.

The Investigator DIPplex Kit has significant advantages over standard STR- and SNP-based DNA profile generation, including the absence of stutter peaks and the very short amplicons (under 160 bp).

Principle

Human identification is commonly based on the analysis of short tandem repeats (STRs) or single nucleotide polymorphisms (SNPs), depending on the demands of an examination or on the sample quality. The Investigator DIPplex Kit uses an alternative means of human identification by allowing the analysis of biallelic deletion/insertion polymorphisms (DIPs, also called INDELs).

Procedure

The 30 biallelic DIP loci plus Amelogenin are simultaneously amplified in a single PCR and labeled with four dyes. See the table "Technical specifications" for more information.

3196 - What is the DIPsorter Freeware?

DIPSorter Freeware has been developed as a utility application for the Investigator DIPplex Kit. Its function is to re-sort genotype data of biallelic deletion or insertion markers — the so-called DIPs.

DIPSorter Freeware enables the import of Genotype Table text files exported from GeneMapper® ID, GeneMapper ID-X, or Genotyper® software and displays them as sorted genotype tables. Generated data can be printed and exported to *CSV and *PDF files.

3194 - What DNA fragment sizes are required for the Investigator DIPlex kit?

The Investigator DIPlex kit produces short amplicon lengths (max. 150 for 30 DIPs) so that DIPplex can be used for identification of very challenging samples (highly degraded).To get a full DIPplex profile, you need DNA fragments of at least 160 bp. If your sample contains fragments of max.75 bp, you will not get any positive results. Usually, degraded samples show varying average fragment sizes. Therefore, a positive result is dependent on the quality of each sample.

3198 - What is the optimal DNA concentration for the Investigator DIPlex Kit?
The optimum DNA input range is between 200-500 pg. The quoted detection limit is 100 pg (full profile with one assay), but reliable results can be obtained with less then 100 pg. For higher amounts of input DNA, consider adjusting the PCR cycle number.
