Principle

为miRNA定量分析选择合适的miScript PCR Control,类似于为mRNA定量分析标准化选择合适的管家基因。对研究中的不同样本类型和不同的处理,有必要验证miScript PCR Control的靶向ncRNA在实验条件下不被调控。另外,要选择表达水平稳定和与miRNA靶基因丰度相似的miScript PCR Control。miScript PCR Control可在GeneGlobe Web门户网站上单独订购。同时提供全套的miScript miRNA PCR Arrays和miScript miRNA QC PCR Arrays。

ΔΔCT法用于相对定量

ΔΔCT法通常用于在miScript PCR System中进行相对定量。该方法主要是比较正常的CT值和实验样本CT值的差异。使用miRNA靶基因的引物(miScript Primer Assay)进行Real-time PCR,同时使用miScript PCR Control检测snoRNA或snRNA。miScript PCR Control得到的循环阈值(CT)用于标准化数据,如下所示。

首先,确定每个样本的循环阈值(CT)。然后,计算ΔCT。ΔCT即miRNA靶基因的CT值与内参ncRNA的CT值之差:

3322 - What is the concentration of the reconstituted miScript Primer Assay (100) and miScript Precursor Assay (100) tubes?

After reconstitution of the lyophilized primers in 550ul nuclease-free water or TE, the miScript Primer Assay and miScript Precursor Assay 100 reaction tubes will be at 10X stock concentration.  They should be used at 1X concentration (0.5 uM) in the PCR/RT-PCR reaction (eg. for a 25ul reaction, 1/10th of the total volume, or 2.5ul, should be primer). For Fluidigm® BioMarkTM System related applications, please refer to the miScript Microfluidics Handbook for primer concentrations and reconstitution recommendations.

Is the expression level of miRNA precursors lower compared to that of mature miRNAs?
In general, precursor miRNA (pre-miRNA) levels are lower than those of mature miRNAs. However this is not always the case. Sometimes the levels of both mature and pre-miRNA may be similar, or pre-miRNA levels may be higher. For this reason, researchers often want to quantify pre-miRNA as well as mature miRNA.  QIAGEN offers miScript precursor miRNA assays as well as mature assays for many miRNAs.
