Product Details

The PAXgene Saliva Collector is a non-invasive sample collection device for human saliva collection, stabilization, transport and storage. The device is designed to simplify at-home collection without the intervention of a lab personnel. The collection device contains a stabilizing solution which maintains the DNA levels in human saliva samples by protecting DNA from degradation and inhibiting bacterial growth over storage time. SARS-CoV-2-derived RNA copy numbers are also stabilized, and the virus is prevented from infecting and replicating in a cell culture model.

The device is part of a comprehensive pre-analytical workflow through nucleic acid extraction and analyses, contributing to reliability and reproducibility of your test results. The PAXgene Saliva Collector is compatible with existing QIAGEN automated and manual extraction kits. The nucleic acids can be used for standard molecular test methods such as RT-qPCR, qPCR, dPCR and NGS.

The PAXgene Saliva Collector is intended for molecular biology applications. This product is not intended for the diagnosis, prevention, or treatment of a disease.

Performance

The PAXgene Saliva Collector is part of a comprehensive pre-analytical workflow for human saliva collection, stabilization, transport and storage through nucleic acid extraction and analyses. The collection device contains a stabilizing solution which stabilizes the DNA levels in human saliva samples by protecting DNA from degradation and inhibiting bacterial growth over time. It is compatible with existing QIAGEN automated extraction kits, such as the QIAsymphony DNA Midi Kit on the QIAsymphony SP Instrument and the QIAamp DNA Mini Kit on the QIAcube (classic and Connect), as well as for manual extraction with the QIAamp DNA Mini and Gentra Puregene Cell Kits.

In addition to DNA stabilization, a research study could show that SARS-CoV-2 (severe acute respiratory syndrome coronavirus type 2)-derived RNA copy numbers are preserved when saliva is collected in the PAXgene Saliva Collector. The viral RNA can be prepared with the QIAamp Viral RNA Mini Kit and quantified with QuantiTect Probe RT-PCR on QIAGEN Rotor-Gene® Q (See " PAXgene Saliva workflow").

Samples collected with the PAXgene Saliva Collector have stable DNA levels for up to 24 months at temperatures up to 25°C. This longevity is possible as DNA is preserved from chemical and enzymatic degradation and bacterial growth is inhibited. After 24 months of storage DNA still retains both high yield and quality if isolated with the QIAGEN DNA extraction kits and protocols specified (See " DNA yield, purity, and integrity is maintained after 24-month storage of stabilized saliva in PAXgene Saliva Collector"). Long-term storage of stabilized saliva at −20 or −80°C is possible (storage study is ongoing).

Protocols and supplementary protocols (see resource section) provide a broad range of existing, proven and flexible QIAGEN DNA extraction solutions for throughput and sample input volume. These protocols provide DNA that is high yield and purity, free of PCR inhibitors.

For manual extraction using silica membrane technology, supplementary protocols are available for the QIAamp DNA Mini Kit spin and vacuum protocols. This kit can also be used for automated DNA extraction on the QIAcube (classic) and QIAcube Connect (See " Automated DNA extraction with QIAamp DNA Mini Kit on the QIAcube (classic) and QIAcube Connect"). Standard protocols for the QIAcube Connect are available on www.QIAGEN.com. Protocols for the QIAcube (classic) can be requested via QIAGEN Technical Support. Optimized protocols for the QIAamp DNA Mini Kit provide high yields of pure DNA, and flexible elution volumes allow DNA concentration to be easily optimized for each downstream application.

The Puregene procedure uses a modified salting-out precipitation method for purification of DNA. The supplementary protocol “Protocol “Purification of DNA from the PAXgene® Saliva Collectors using the Gentra® Puregene® Cell Kit” for the Gentra Puregene Cell Kit offers a simple solution for manual DNA extraction from different sample input volumes, including the whole content of a PAXgene Saliva Collector (See " Consistently high DNA yield and purity with flexible sample input volume when using the Gentra Puregene Cell Kit").

Medium- to high-throughput automated purification of DNA from the PAXgene Saliva Collector is possible on the QIAsymphony SP Instrument using protocols for the QIAsymphony DNA Midi Kit for sample volumes of 400 and 1000 µl. High reproducibility is guaranteed through standardized magnetic bead processing technology (See " Automated DNA extraction using the QIAsymphony DNA Midi Kit on the QIAsymphony SP Instrument.").

When using either one of the aforementioned QIAGEN DNA extraction kits, the obtained saliva gDNA is not only of high purity but also of high integrity (see " Human Genomic DNA obtained from PAXgene Saliva samples is of high molecular weight." part A) The DNA profile is maintained during long-term storage of two years at room temperature (15-25°C) (see " Human Genomic DNA obtained from PAXgene Saliva samples is of high molecular weight." part B).

Stabilization characteristics have been shown to be robust against elevated levels of endogenous and potentially interfering substances, under- or overfilling and inappropriate mixing (see technical note ´PAXgene Saliva Collector Robustness Study´ in the resource section). DNA isolated with the specified QIAGEN DNA extraction kits and protocols is free from PCR inhibition and is compatible with downstream analytical PCR assays, including digital PCR with the QIAGEN QIAcuity Digital PCR system (See " Absolute Quantification of Gene Copies by dPCR using the QIAcuity Digital PCR system"), and next-generation sequencing (NGS) molecular test methods.

In addition to the genomic DNA, a research study could show that the SARS-CoV-2 (severe acute respiratory syndrome coronavirus type 2) is stabilized in saliva collected with the PAXgene Saliva Collector for at least 4 days (96 hours) at 20°C. The genome of SARS-CoV-2 consists of RNA and can be isolated from the saliva with QIAamp Viral RNA Mini Kit for quantification with QuantiTect Probe RT-PCR and SARS-CoV-2 N1+N2 Assay Kits on QIAGEN Rotor-Gene Q ( See " PAXgene Saliva stabilizing solution maintains SARS-CoV-2 derived RNA levels for at least 96 h and reduces infectivity of SARS-CoV-2" part A).

While the PAXgene Saliva Collector stabilizes SARS-CoV-2 particles for detection and quantification it efficiently inactivates the virus, rendering it unable to infect and replicate in a cell culture model (See " PAXgene Saliva stabilizing solution maintains SARS-CoV-2 derived RNA levels for at least 96 h and reduces infectivity of SARS-CoV-2" parts B, C, D).

Principle

The PAXgene Saliva workflow starts with the collection of human Saliva in PAXgene Saliva Collectors containing a stabilizing solution.

The PAXgene Saliva Collector is a plastic device with a funnel screwed to a collection tube which enables 2.0 ml human saliva collection. The funnel consists of an inner cavity containing 1.0 ml of the stabilizing solution. The inner cavity is secured to the inner collection tube which helps prevent spillage of the solution (See " Components of the PAXgene Saliva Collector"). The stabilizing reagent is released upon unscrewing of the funnel (See " Instructions for use of the PAXgene Saliva Collector"). The stabilizing agent has bacteriostatic properties which inhibit bacterial growth over storage time. It is free of crosslinking, or nucleic-acid modifying substances.

See figures

Procedure

In contrast to invasive specimen collection, saliva collection with the PAXgene Saliva Collector does not require trained professionals or dedicated facilities to collect a saliva sample. With clear instructions and verified collection device safety claims, saliva specimens can be easily self-collected with the PAXgene Saliva Collector from most people at home or outside of a doctor´s office.

Saliva is spit into the funnel until the liquid saliva (excluding bubbles) fills up outer part of the tube and reaches the fill line. This usually takes about 5−10 min. When the fill line is reached, the funnel is slowly unscrewed from the tube with the tube still held in upright position. This releases the stabilization solution from the inner part of the tube. After complete liquid transfer the funnel is discarded, a lid is removed from the bottom of the tube, placed over the opening and screwed tightly to close the tube. Finally, the tube is inverted 5 times to mix saliva and stabilizing solution (See " Instructions for use of the PAXgene Saliva Collector").

For transportation of the stabilized saliva sample, the PAXgene Saliva Collector tube can be put back into the blister box without the funnel.

3828
What happens if I accidentally unscrew the funnel before saliva collection is completed?

Stabilizing reagent included in the funnel extension will leak into the collection tube. As a consequence, correct filling will not be possible anymore.

3808
May I continue with the saliva collection if I accidentally unscrewed the funnel prematurely?

If the stabilizing reagent leaked into the collection tube before saliva collection was completed, the fill line does not indicate the correct volume of saliva anymore.
We therefore recommend in this case to repeat saliva collection with a new device.

3809
Is it possible to place the PAXgene Saliva Collector tube directly on the QIAsymphony SP instrument sample rack?

No. Stabilized saliva from the PAXgene Saliva Collector tubes have to be transferred into a secondary tube.
See labware list for more detailed information available under the QIAsymphony DNA Midi Kit product page on www.QIAGEN.com.

3825
What can I do to stimulate saliva flow?

Massage your cheeks with both hands for 15–30 seconds.

3824
What is the expected DNA yield per milliliter saliva collected and stabilized with the PAXgene Saliva Collector?

DNA yield can differ between individual donors by up to 20-fold. Median human DNA yield, as determined by qPCR, from 290 different sample preparations with the Gentra Puregene Cell Kit was 16 µg DNA per ml saliva.

3822
Which QIAGEN DNA extraction kits are compatible with the PAXgene Saliva Collector?

Protocols for automated DNA extraction from saliva stabilized with the PAXgene Saliva Collector are available for the QIAsymphony DNA Midi Kit with the QIAsymphony SP instrument or the QIAamp DNA Mini Kit with the QIAcube (Classic and Connect).
For manual extraction, supplementary protocols are available for the QIAamp DNA Mini and Gentra Puregene Cell Kits (see resources section).

3821
How long does a saliva collection take?

Most users take up to 5–10 minutes to complete collection of 2 ml saliva.

3804
What should I avoid before sample collection?

Avoid eating, drinking, chewing gum, kissing, and smoking 30 minutes prior to sample collection. No dental work should be performed within 24 hours prior to sample collection. We recommend rinsing your mouth 30 minutes prior to collection to remove any food residues.

3813
How should unused PAXgene Saliva Collector be stored?

Store the unused PAXgene Saliva Collector within the blister box at room temperature (15–30°C).

3812

3829
Can saliva in PAXgene Saliva Collector tubes be stored frozen at –20 or –80°C?

Yes. Saliva collected into PAXgene Saliva Collector can be stored frozen. For latest results on storage at ambient or frozen temperature, see the technical notes at the resource section of www.QIAGEN.com and  www.PreAnalytiX.com.

For storage at –20 or –80°C, it is recommended to transfer the sample into a suitable cryovial.

3817
Which downstream applications have been tested with DNA purified from saliva collected into PAXgene Saliva Collector?

DNA from saliva samples collected using the PAXgene Saliva Collector have been successfully tested in PCR, including multiplex qPCR, dPCR, and NGS applications.

3827
How does the stabilizing reagent in the PAXgene Saliva Collector stabilize saliva?

The stabilizing reagent in the tube prevents microbial growth and chemical as well as enzymatic degradation of DNA in saliva.

3814
Are pathogens inactivated in human saliva samples collected with the PAXgene Saliva Collector?

Specimens and samples are potentially infectious and must be treated as biohazardous materials.
Evidence exists that SARS-CoV-2 is inactivated in the stabilizing reagent in a way that the virus loses its capability to replicate in a cell culture model (for more information see the application note at the Resource section of www.QIAGEN.com and  www.PreAnalytiX.com).

3819
What is the expected DNA purity from saliva collected and stabilized with the PAXgene Saliva Collector?

Total DNA purified from the PAXgene Saliva Collector with the QIAGEN DNA purification kits has A260/A280 ratios of 1.7–1.9, and absorbance scans show a symmetrical peak at 260 nm.

3823
Is it possible to purify DNA from the whole sample of a PAXgene Saliva Collector in one preparation?

Yes. A supplementary protocol for DNA extraction of 3 ml sample (2 ml saliva plus 1 ml stabilizing reagent) is available for the Gentra Puregene Cell Kit (for the supplementary protocol, see Resources section).

3826
What can I do if I don't see the fill line anymore due to saliva bubbles?

Wait until bubbles disappeared and fill line is visible again.

3807
Can saliva in PAXgene Saliva Collector tubes be stored at temperatures higher than 25°C?

DNA profile remains stable through short-term exposure to elevated temperatures up to 50°C.

3816
Can I stop saliva collection before the fill line is reached?

The PAXgene Saliva Collector is robust against underfilling. However, only a complete fill ensures optimal results. Reduced volume of saliva will lead to lower nucleic acid yield, which might negatively impact subsequent molecular analyses.

3806
What is in the additive in the PAXgene Saliva Collector?

The PAXgene Saliva Collector contains 1 ml of a liquid stabilizing reagent with a proprietary formulation that prevents microbial growth and DNA degradation in saliva.

Contents of the tube may cause irritation to the eyes, respiratory system, and skin. Please refer to the PAXgene Saliva Collector Handbook, accessible under the Resources tab on www.PreAnalytiX.com, for more details. Additional information can be found in the Safety Data Sheet, also accessible under the Resources tab.
