原理

QIAGEN Vacuum Pump适用于实验室标准真空操作（34 L分钟, 8 mbar）。Vacuum Pump配套QIAvac真空底座，可真空处理QIAGEN的多种核酸纯化试剂盒。在真空的作用下，液体流过，可替代离心操作，减少样本制备所需的手动操作时间。

QIAvac Holder（参见" QIAvac Holder"）可与Tube Extenders用在QIAvac 24 Plus上，放置QIAGEN Plasmid Plus Mega and Giga离心柱，防止离心柱晃动，尤其在离心柱含有大体积液体时。

VacValves可直接插在QIAvac 24 Plus底座的鲁尔接头上，应在样本流速有显著差异时使用。在处理粘稠样本，如血液时，应使用VacValves以确保真空流速均匀。

VacConnectors（参见" VacConnector"）是一次性接头，可装在QIAamp离心柱或QIAprecipitator模块的外管口，也可接在QIAvac 24、QIAvac 6S（已停产）的QIAvac Luer Adapters或QIAvac 24的VacValves的鲁尔接头上。该配件可代替离心，用于分离核酸。VacConnectors可防止在纯化过程中离心柱或模块与鲁尔接头直接接触，避免产生任何交叉污染。VacConnectors为一次性产品，使用后应丢弃。

QIAvac Luer Adapters可在QIAvac 6S（已停产）上便利的处理至多24个离心柱。在真空作用下，样本和洗涤液流过离心柱，因此无需离心，加快了纯化过程。QIAprep、QIAquick和MinElute离心柱的接口可直接插在QIAvac Luer Adapters的鲁尔接头上。每个Luer Adapter含有4个鲁尔接头（参见照片），QIAvac 6S上层板上可连接6个Luer Adapters，因此一次可处理24个样本。QIAamp离心柱在QIAvac 6S（已停产）上使用时需配套使用VacConnectors和QIAvac Luer Adapters。

Vacuum Regulator（参见" Vacuum Regulator"）可便利的监控并调整使用QIAvac底座时的真空压。QIAGEN纯化试剂要求真空压在一定范围内（如产品使用手册中所写），这样才能获得理想产量和理想纯度的核酸。

The procedure has been used successfully for isolation of a single-copy, 14.5 kb, binary plasmid, p35S GUS INT, from Agrobacterium tumefaciens strain GV2260 (1).

This procedure has been adapted by customers and is for purification of viral RNA and DNA from plasma, serum, and cell-free body fluids using the QIAamp MinElute Virus Vacuum Kit.

We do not recommend using a vacuum manifold for DNA extraction using the QIAamp 96 DNA Blood Kit because:

Viscosity of the lysates varies from sample to sample. Therefore, the wells of the QIAamp 96 plate are at great risk of clogging.
Use of vacuum manifold would result in incomplete removal of ethanol and salts after washing with Buffer AW2. This will affect the purity and yield of samples.

It is possible to use a general laboratory vacuum system with the QIAamp DSP Virus Kit. However, this will require validation in-house. The CE-marked QIAamp Kits have been developed, field-tested and validated with the QIAvac 24 Plus vacuum system (QIAvac 24 Plus, QIAvac Connecting System and Vacuum Pump). Using this system with the QIAamp DSP Virus Kit will avoid lengthy validation procedures for the enduser.

QIAvac Multiwell can hold plates that conform to SBS plate standards, whereas the QIAvac 96 does not. Therefore, the waste tray for QIAvac Multiwell is not the same as for our QIAvac 96.

However, the waste tray from QIAvac Multiwell will fit the QIAvac 96, but not vice versa.
