原理

therascreen KRAS RGQ PCR Kit用于检测KRAS基因的12和13位密码子的突变。KRAS基因编码的蛋白在EGFR信号通路中发挥重要作用。KRAS基因的突变会影响蛋白刺激下游通路。KRAS基因在各种癌症类型中约30%会发生突变。高频率KRAS突变的癌症包括结直肠癌(35%)和肺癌(18%)。

    试剂盒利用两种技术,即ARMS(Amplification Refractory Mutation System)和Scorpions,通过real-time PCR检测突变。

    ARMS

    通过ARMS可进行等位基因或突变特异性扩增。Taq DNA聚合酶可有效区分PCR引物3'端的搭配与错配。特定突变序列被选择性扩增,即使样本中大部分序列都不含有此突变。当引物完全搭配时,扩增过程非常高效。当3'端碱基错配时,只有低水平背景扩增发生。

    Scorpions

    使用Scorpions检测扩增。Scorpions是双功能分子,含有一个共价连接到探针上的PCR引物。探针上的荧光基团与同样在探针上的淬灭基团相互作用,降低了荧光强度。PCR过程中,当探针结合到扩增子上时,荧光基团和淬灭基团就会分开,导致反应管中荧光强度增加。

    What technologies are used by the therascreen PCR kits?
    The mutations in the therascreen PCR kits are detected with high sensitivity using the Amplification Refractory Mutation System (ARMS). PCR is used to amplify the mutation, and amplification is signalled using Scorpions technology. Further information on these technologies can be obtained from QIAGEN Technical Service.
